Scientists revolutionize creation of genetically altered mice to model human disease

May 2, 2013 ? Whitehead Institute Founding Member Rudolf Jaenisch, who helped transform the study of genetics by creating the first transgenic mouse in 1974, is again revolutionizing how genetically altered animal models are created and perhaps even redefining what species may serve as models.

"This new method is a game changer," says Jaenisch, who is also a professor of biology at MIT. "We can now make a mouse with five mutations in just three to four weeks, whereas the conventional way would take three to four years. And it's rather straightforward, probably even easier than the conventional way."

Scientists create models in mice by altering specific genes that have been associated with a given disease. The models allow for the study of the development and course of the disease and the effects of various interventions, including genetic and chemical. For the past 20 years, the creation of such models has remained relatively unchanged: scientists insert a piece of DNA into a mouse embryonic stem (ES) cell, inject the modified cell into a very early-stage embryo, called a blastocyst, then implant this developing ball of cells into a foster female mouse. The whole process can take years and tens of thousands of dollars to establish a mouse strain with, for example, a single copy of a gene "knocked out." Such knockouts can only be created in very few species, including mice and rats, whose ES cells can be grown and modified reliably.

The new approach used by scientists in Jaenisch's lab bypasses ES cells to quickly and efficiently produce mice with mutations in both copies of multiple genes. In next week's issue of the journal Cell, Haoyi Wang, Hui Yang, and Chikdu Shivalila describe their technique, which is based on a system that certain bacteria use to fend off viral attacks.

This is the first time that the system, known as CRISPR (for "clustered regularly interspaced short palindromic repeat")/Cas (for "CRISPR-associated"), has been used to alter multiple genes in a single multicellular organism. Shivalila says the process is so accessible that he expects other labs to adopt it quickly.

"For any institution or university with a core facility, we think this will be the way they will start making mice carrying specific mutations because it's a lot faster and so efficient," says Shivalila, one of Jaenisch's graduate students. "We were surprised that we could get two genes 'knocked out' at four loci very, very efficiently, about 80% efficiency. If we used TALENs, a more recent and complicated development in genetic engineering, we got 30% efficiency for just one gene."

Because the CRISPR/Cas technique can generate mutant mice even without using ES cells, a limitation of the conventional method for making models, genetic research may no longer be confined to a limited list of model organisms -- those for which ES cells exist.

"This breaks down the definition of model organism," says Wang, a postdoctoral researcher in Jaenisch's lab. "So now, even with limited resources, any animal with established embryo manipulation procedures could be the subject of genome engineering. With many of the animals' genomes that have been sequenced, we could use this technology to establish efficient genetic manipulations in more species, to study the unique biology of each, and to learn more about evolution."

Thus, Wang, Yang, and Shivalila have used CRISPR/Cas to create mouse models only, but the team is excited broaden its application to other animals.

"We also need to see if the CRISPR/Cas system has any unexpected, undesired off-target effects, changes to the genome that we don't want," says Yang, a postdoctoral researcher in the Jaenisch lab. "So we need study this further to establish the fidelity of the system. But I think this will be the way to go."

Would arming Syria's rebels just prolong the war?

And would it be enough to tip the balance of power against Bashar al-Assad?

With evidence mounting that Syria's armed forces have started using chemical weapons against rebels, the Obama administration is reportedly considering getting more deeply involved in the country's bloody civil war by sending arms to some of the opposition fighters. Republican senators are putting pressure on President Obama to act fast, saying that the move is long overdue. But a key question persists: After more than two years of fighting and 70,000-plus deaths, would giving the rebels greater power really give them the boost they need to topple President Bashar al-Assad's regime?

Some analysts say the answer is no. Obama is reportedly considering supplying rebels with everything from communications gear to armored vehicles to shoulder-fired anti-aircraft missiles. "That military gear can prolong the conflict, preventing dictator Bashar Assad from crushing the rebels," says Spencer Ackerman at Wired. "It is unlikely to tip the balance of the war toward the rebels so they can decisively" win.

Assad has a variety of advantages ? an adaptive military estimated at over 50,000; complete air superiority; chemical weapons ? that he will retain even if Obama opens a new arms pipeline. Overcoming those advantages means getting, at the least, U.S. and allied airpower involved ? a step the Obama administration, and especially the military, want to avoid. Especially since it might involve shooting down Iranian planes, a fateful step. [Wired]

It's still worth a shot, others say. Early in Syria's uprising, "a sharp diplomatic and economic shove might have convinced elements of the regime to give up on the Assad family," says Michael Gerson at The Washington Post. But Obama wanted to avoid wading into another conflict in the Middle East, in hopes of "improving the image of America in the Islamic world." Things obviously didn't pan out. Now he's "missed the moment," and the best we can hope for is limiting the damage we could face from "a civil war at the heart of the Middle East that destabilizes friendly governments, empowers jihadists, increases sectarian tensions across the region, and allows Iran broader opportunities for mischief."

The United States should not accept the refounding of the Assad regime on a vast pile of skulls. And the United States should not accept the disintegration of Syria into enclaves, some of them sheltering al-Qaeda affiliates. So Obama has little choice but to help the rebels to win, while helping the right rebels to win out in a post-Assad power struggle. But delay has made this task much more difficult. Civil wars, over time, tend to favor the ruthless and radical. As options for arming the rebels narrow ? for fear of weapons falling into the hands of future enemies ? the pressure for direct U.S. action (particularly a no-fly zone) increases. [Washington Post]

One nearly certain outcome is that beefing up the rebels' arsenal would escalate the violence. "It would also make the regime of Bashar Assad more desperate," says Fareed Zakaria at TIME. "Perhaps Assad has already used chemical weapons; with his back against the wall, he might use them on a larger scale."

If the objective is actually to reduce the atrocities and minimize potential instability, the key will be a political settlement that gives each side an assurance that it has a place in the new Syria. That was never achieved in Iraq, which is why, despite U.S. troops and arms and influence, the situation turned into a violent free-for-all. If some kind of political pact can be reached, there's hope for Syria. If it cannot, U.S. assistance to the rebels or even direct military intervention won't change much: Syria will follow the pattern of Lebanon and Iraq ? a long, bloody civil war. And America will be in the middle of it. [TIME]

HTC marketing blitz to squeeze 2013 margins

TAIPEI (Reuters) - Taiwanese smartphone maker HTC Corp will struggle to improve its margins this year as it increases marketing spend for its latest model in a bid to catch up to rivals Samsung Electronics Co Ltd and Apple Inc.

HTC on Thursday forecast strong sales of its flagship HTC One smartphone to lift second-quarter revenues by almost two-thirds to T$70 billion ($2.37 billion).

Operating margins were also expected to widen to between 1 and 3 percent from the previous quarter's 0.1 percent, as HTC spends more on a campaign to improve its brand image. HTC's third-quarter 2012 operating margins were 7 percent.

"We're improving the HTC marketing execution. It's the first time since HTC developed its brand that we are really integrating brand, product and marketing all together," HTC Chief Executive Peter Chou told an investor conference call.

He did not give specific figures but the company said in March its digital media marketing budget for the first half of this year would increase 250 percent year-on-year, while the budget for traditional media will double.

Investors, however, were cautious about the HTC One's longer-term prospects as both Apple and Samsung have much deeper pockets and are more established brands.

"Samsung basically cornered the whole supply chain. To me, HTC is more of a product story: good product, higher stock price," said Hong Kong-based portfolio manager Michiel Van Voorst, who manages $1.2 billion in Asian equities for Robeco.

"I think investors will be a bit more reluctant now to price the stock at earnings that have not yet materialized," added Van Voorst, who cut holdings in HTC after its fourth-quarter 2012 profit missed forecasts and plunged 91 percent year-on-year.

The HTC One, currently available in five countries including China, runs a version of Google Inc's Android software that allows users to customize their homescreen.

Research firm Daiwa Capital, in a recent report, said HTC One was one of the top-five best selling smartphones offered by U.S. carriers and that it was out of stock at some operators and retailers in Britain, Germany and Taiwan.

KGI Securities analyst Richard Ko believes this popularity may be short-lived.

"While we think HTC's shipments will rebound strongly in Q2, we also think Samsung and other second tier players will continue to exert pressure on HTC," he said. "We remain cautious because of HTC's execution capability and long-term structural weaknesses."

Delays in the full launch of the HTC One due to a shortage of a camera component led HTC to report a record-low net profit in the previous quarter.

Future sales volumes were also thrown into doubt after rival Nokia Oyj won a court injunction last week that would prevent HTC from using microphone components made by STMicroelectronics NV in HTC One phones.

HTC has said it does not expect the decision to have any immediate impact on sales, and that it will find other suppliers once its inventory of STMicroelectronics microphones runs out.

HTC was the world's 10th-biggest smartphone maker by shipments in the fourth quarter, according to IT research firm Gartner, jostling in a crowded field behind Samsung and Apple.

Shares in HTC have slipped 1.8 percent this year, underperforming the broader market's 5.6 percent rise. On Thursday, HTC shares closed down 2 percent compared to a 0.43 percent rise in the main share index.

Misperceptions Keep Seniors from Seeking Credit Counseling ...

When it comes to financial counseling, a number of consumer misconceptions prevent them from seeking the proper help, according to a recent survey from the National Foundation for Credit Counseling (NFCC).

The survey revealed several areas of confusion associated with credit counseling, such as where to turn for such assistance, the cost of counseling and the idea that credit counseling would end up hurting individuals? credit scores.?

NFCC attempts to clear the air of any confusion consumers might have by listing several facts regarding credit counseling services.?

While it is understandable to be cautious when dealing with financial matters, NFCC urges consumers to look for agencies associated with membership organizations as well as checking with the Better Business Bureau and state Attorney General?s office to see if there are any unresolved complaints against any agency they are considering.

Another anxiety NFCC noticed among its respondents was a concern about the cost of counseling, many of which believing it costs ?too much.?

Others believed counseling would hurt their credit score, for fear that counseling agencies report their individual scores to credit bureaus after the session is complete.?

The idea that counseling agencies only offered advice and not real solutions was another concern survey respondents admitted.?

To dispel this misconception, NFCC reassures that the goal of the counselor is to provide both short-term and long-term solutions for consumers.?

Written ?action plans? and ?debt management plans? are tools that can be used to help financially struggling persons meet debt obligations and lay the groundwork for a financially stable tomorrow.?

?When struggling with debt and other personal finance issues, consumers do themselves and their families a disservice by not seeking advice from a trusted professional nonprofit credit counseling agency,? said Gail Cunningham, spokesperson for the NFCC.?

There can be major differences among agencies, says Cunningham, who assures that NFCC member agencies are required to be accredited by an independent third party and all counselors must become certified in order to provide counseling services.

Manufacturing activity declines in April

Manufacturing activity declined in April, according to the Institute for Supply Management's latest Report on Business for the manufacturing sector.

At 50.7 the purchasing manager?s composite index (PMI) declined 1.17 percent since March 2013 and dropped 6.28 percent below the level seen a year earlier. That's an indication of slowing manufacturing activity for the second consecutive month.

Zinc: The Goldilocks metal for bioabsorbable stents?

Apr. 30, 2013 ? In 2012, more than 3 million people had stents inserted in their coronary arteries. These tiny mesh tubes prop open blood vessels healing from procedures like a balloon angioplasty, which widens arteries blocked by clots or plaque deposits. After about six months, most damaged arteries are healed and stay open on their own. The stent, however, is there for a lifetime.

Most of the time, that's not a problem, says Patrick Bowen, a doctoral student studying materials science and engineering at Michigan Technological University. The arterial wall heals in around the old stent with no ill effect. But the longer a stent is in the body, the greater the risk of late-stage side effects. For example, a permanent stent can cause intermittent inflammation and clotting at the implant site. In a small percentage of cases, the tiny metal segments that make up the stent can break and end up poking the arterial wall.

"When the stent stays in place 15, 20 or 25 years, you can see these side effects," says Bowen. "It's not uncommon to have a stent put in at age 60, and if you live to be 80, that's a long time for something to remain inert in your body."

That's why researchers are trying to develop a bioabsorbable stent, one that would gradually -- and harmlessly -- dissolve after the blood vessel is healed.

Many studies have investigated iron- and magnesium-based stents. However, iron is not promising: it rusts in the artery. Magnesium, on the other hand, dissolves too quickly. "We wondered, 'Isn't there something else?'" Bowen said. "And we thought, 'Why not zinc?'"

So they placed tiny zinc wires in the arteries of rats. The results were amazing. "The corrosion rate was exactly where it needed to be," Bowen said. The wires degraded at a rate just below 0.2 millimeters per year -- the "magic" value for bioabsorbable stents -- for the first three months. After that, the corrosion accelerated, so the implant would not remain in the artery for too long. On top of that, the rats' arteries appeared healthy when the wires were removed, with tissue firmly grasping the implant.

"Plus, zinc reduces atherosclerosis," he added, referring to zinc's well-known ability to fight the development of plaque in the arteries. "How cool is that? A zinc stent might actually have health benefits."

There is one drawback. "A stent made of conventional zinc would not be strong enough to hold open a human artery," he said. "We need to beef it up, double the strength."

"The good news is that there are commercial zinc alloys that are up to three times stronger," Bowen said. "We know we can get there. We just don't want to ruin our corrosion behavior."

The researchers have filed a provisional patent on their discoveries and are now testing new zinc-based stent materials.

An article on their work, "Zinc Exhibits Ideal Physiological Corrosion Behavior for Bioabsorbable Stents," was recently published in the journal Advanced Materials.

Bowen's research is supported by a two-year, $52,000 predoctoral fellowship from the American Heart Association. Initial research was supported by a summer fellowship from the DeVlieg Foundation.

Bowen's advisor is Jaroslaw Drelich, a professor of materials science and engineering, and they work in close collaboration with Jeremy Goldman, an associate professor of biomedical engineering. Undergraduates Jacob Braykovich and Matt Tianen are also working on material development and corrosion testing related to the project.
